Murashige & Skoog (MS) Medium, developed in 1962, is the most widely used plant tissue culture medium. It was originally formulated for the cultivation of Nicotiana tabacum callus and evolved from earlier media such as White’s medium with significantly increased nutrient concentrations. The formulation provides high levels of macro- and micronutrients, particularly nitrogen (typically 50–60 mM), which strongly promotes cell proliferation and callus growth. Higher nitrogen levels (>80 mM) were found to be inhibitory, highlighting the importance of balanced nutrient composition. Due to its relatively high salt strength, MS medium is considered a nutrient-rich medium and may be too concentrated for certain sensitive species. For this reason, modified forms such as half-strength or reduced macronutrient MS are commonly used. MS medium serves as a universal baseline for plant tissue culture systems.
